Recombinant monoclonal antibody to Hexon. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma 9C12.

Immunogen: The original antibody was generated by immunizing mice with Ad type 5 (Ad5).
Specificity: The antibody binds hexon protein from Ad5 particles. In particular, it binds to HVRs 2, 5 and 7 of the hexon. Hexon is the major coat protein of adenovirus.
Application Notes: The antibody was able to neutralize Ad5. Cryo-EM reconstruction of the Ad5-9C12 complex showed that the capsid is coated by a meshwork of bivalently bound antibody molecules. The antibody bound to the hexon protein of Ad5 particles denatured as shown by western blot analysis (Varghese et al., 2004; PMID: 15507619).The crystal structure of scFv fragment of the antibody in complex with human adenovirus C5 hexon was determined (Schmid et al., 2018; PMID: 29386504). The specificity of the original format of the antibody was confirmed by ELISA analysis (Moyer et al., 2016; PMID: 26491163). Wild-type viruses were immunoprecipitated using the antibody (Myerset al., 2013; PMID: 23487468). The binding affinity of the Fab fragment to Adv5 hexon was measured by surface plasmon resonance (Kd= 1 nM) (Bottermann et al., 2016; PMID: 27881870). The antibody detected hexon on A549 cells by immunofluorescence analysis (Smith et al., 2008; PMID: 18448546).
