Recombinant monoclonal antibody to kappa light chain. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma NH3/41.34.

Immunogen: This antibody was raised by immunising Balb/c mice with whole IgG fraction and subsequent hybridoma cells formation.
Specificity: This antibody recognises human immunoglobulin kappa light chain. The antibody does not cross react with rhesus macaque IgG1, cynomolgus monkey IgG1 and cynomolgus monkey IgG4.
Application Notes: NH3/41.34 antibody detects kappa light chains from a human IgGs and Bence-Jones protein samples via Western blot under reducing conditions (Thorpe et al., 1984). This antibody is equally able to detect full IgGs via Western blot when the samples are transferred from the non-reducing SDS polyacrylamide gels (Thorpe et al., 1984). Further, this antibody was capable of detecting IgGs via Western blot from native gels run under acidic conditions (Thorne et al., 1984). Finally, the specificity of the antibody for human IgG1 was confirmed by ELISA analysis. No cross reactivity with rhesus macaque IgG1, cynomolgus monkey IgG1 and cynomolgus monkey IgG4 was detected.

ELISA of anti-kappa light chain (human) antibody on human kappa light chain-Fc fusion protein. Binding curves of the rabbit IgG chimeric version of the anti-kappa light chain (human) antibody NH3/41.34 (Ab00855-23.0; red line) and isotype control (Ab00102-23.0, anti-fluorescein; blue line) to an ELISA plate coated either with Pr00115 (kappa light chain dimer protein; A) or Pr00116 (lambda light chain dimer protein; B) each at a concentration of 5 µg/ml. A 3-fold serial dilution from 10,000 to 0.17 ng/ml of Ab00855-23.0 was tested in each experiment. For signal detection, a 1:4000 dilution of a HRP-conjugated anti-rabbit IgG1 antibody was used. Ab00895-23.0 bound Pr00115 well, but some background reactivity of Ab00102-23.0 with Pr00115 was observed at high antibody titres. Ab00855 does not react with Pr00116.

A 3-fold serial dilution from 3000 to 0.0169 ng/ml was performed using Ab00854-1.1 and an anti-mouse IgG was used as secondary antibody for detection.
