Immunogen: The original antibody was selected from a commercially available phage display library. The library was panned against the receptor-binding domain of human M-CSF (rbdM-CSF).
Specificity: This antibody is specific for human M-CSF. It has been shown to cross-react with cynomolgus monkey M-CSF. M-CSF is a secreted cytokine that influences hematopoietic stem cells to differentiate into macrophages or other related cell types.
Application Notes: The original version of this antibody (human IgG1) was tested for its specificity for human M-CSF and cross-reactivity for cynomolgus M-CSF using ELISA. KD values of 14 pM and 16 pM were measured. This antibody binds M-CSF expressed on cells, as shown in a FACS assay that exhibited an EC50 value of 9.8 nM and 14 nM against M-CSF-expressing transfected CHO cells and MDA-MB 231 cells, respectively. This antibody successfully inhibited human and cynomolgus M-CSF binding to recombinant human M-CSF receptor with an IC50 of 6.1 pM and 6.3 pM, respectively. This antibody successfully inhibited the proliferation of M-NFS-60 cells induced by recombinant and native human and cynomolgus M-CSF with an IC50 of 3.1 pM and 17.7 pM for recombinant and native human M-CSF, respectively, and 4.3 pM and 106.9 pM for recombinant and native cynomolgus M-CSF, respectively. This antibody successfully inhibited the proliferation of M-NFS-60 cells induced by membrane-bound human M-CSF (expressed by stably transfected CHO cells) (US10081675B2).
