Specificity: This antibody binds the human NKG2a. It can also recognize and bind the CD94/NKG2a heterodimer formed by the covalent association of CD94 with the NKG2a molecule. The CD94/NKG2a heterodimer acts as an inhibitory natural killer cell receptor and binds nonclassical MHC class I protein, HLA-E. NKG2a is found mainly on NK cells and on subsets of CD8+ cells.
Application Notes: This antibody was reported to be a non-competitive antagonist of human CD94/NKG2A receptor expressed on lymphocytes, since it inhibited the function of CD94/NKG2A. Z199 can block the function of human CD94/NKG2A, and specifically induce killing of cells by CD94/NKG2A-restricted lymphocytes in a concentration-dependent fashion (US8796427). This antibody was used to study the covalent interactions between CD94 and NKG2A, which together form the inhibitory natural killer cell receptor (PMID: 9045931). This antibody was used to study the binding interactions of HLA-E (wild-type) tetramer to CD94/NKG2A or CD94/NKG2C expressing cells by flow cytometry (PMID: 14971033). This antibody was used to test the cytotoxic activity of NKL cells against tumor cells. NKL cells were tested for their ability to lyse the indicated targets in a 51Cr release assay, this antibody was found to enhance NK lysis (PMID: 21969815).
