Immunogen: The original antibody was generated by immunizing Lama glama with recombinant N protein (prNΔ85) of a Brazilian hantavirus strain. The VHH regions were isolated to construct an immune library. VHHs were displayed fused to the M13KO7 phage coat protein III and the selection steps were performed on immobilized prNΔ85.

Specificity: This antibody specifically binds prNΔ85 of the Araucaria hantavirus protein N. Hantaviruses are rodent-borne RNA viruses that belong to the Bunyaviridae family and can cause Hemorrhagic Fever with Renal Syndrome (HFRS), more commonly found in the Old World, and Hantavirus Pulmonary Syndrome (HPS), present mostly in the American continent. HPS is transmitted to humans by inhalation of virus-contaminated aerosols from rodent excreta.

Application Notes: The binding specificity of this antibody for recombinant prNHΔ85 protein of Araucaria hantavirus was determined using ELISA. This antibody can also bind prNΔ85 in a western blot. The binding affinity of this antibody for immobilized prNΔ85 was determined using SPR. The binding affinity of the original VHH nanobody was reported to be Kd= 38.9 nM. This antibody can detect prNΔ85 in solution in an ELISA, as well as the native viral antigen from rodent serum sample (PMID: 25243411).
