Recombinant monoclonal antibody to Oligodendrocyte-specific surface antigen. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma SCH94.03.

Immunogen: The original antibody was generated by immunizing SJL/J mice with spinal chord homogenate (SCH) in incomplete Freund's adjuvant.
Specificity: This antibody recognized an antigenic epitope found on the surface of oligodendrocytes. This antibody reacts with all major organs including the brain, spinal cord, heart, liver, kidney, stomach, and small intestine (PMID: 7931575).
Application Notes: The original IgM version of this antibody was found to promote remyelination of the central nervous system when passively transferred to SJL/J mice chronically infected with Theiler's murine encephalomyelitis virus (TMEV). The screening of the hybridomas for significant binding towards SCH was done using ELISA. Immunohistochemistry using this antibody showed multiorgan autoreactivity, and that it recognized both surface and cytoplasmic determinants on glial cells. The characterization of the antigen recognized by this antibody, various cell lines derived from glial (rat C6, mouse G26-20, human U373MG and U87MG), neural (human neuroblastoma), fibroblast (mouse L and 3T3), epithelial (human SCC-9 carcinoma), and lymphocytic (mouse CTLL2) origin were stained. This antibody reacted with internal antigens of all the cell lines tested (PMID: 7931575). A treatment with this antibody reduced by 2- to 3-fold the number of CD4(+) and CD8(+) cells infiltrating the CNS of SJL/J mice chronically infected with TMEV, in the absence of global lymphocyte depletion. Immunohistochemical staining showed that SCH94.03 labeled MHC class II-positive dendritic cells in peripheral lymphoid organs (PMID: 8671597). To elucidate the mechanism of antibody-mediated remyelination, this antibody was investigated immunocytochemistry, flow cytometry, immunoelectron microscopy, Western blotting, and immuno-thin layer chromatography. In western blotting of secondary isolated oligodendrocytes lysates, this antibody showed reactivity with multiple protein bands of 27, 32, 50, 100, and 106 kDa (PMID: 8714516).
