Immunogen: The original antibody was generated by immunizing BALB/c mice intraperitoneally with 5 to 10 micro g purified native human rhinovirus 2 emulsified in complete Fruend's adjuvant.
Specificity: This antibody specifically binds a linear neutralizing epitope on the human rhinovirus type 2, between amino acid residues 153 and 164 of virus capsid protein VP2. Later, the minimal binding site of 8F5 wasa mapped to a sequence 'TRLNPD' covering VP2 residues 160 to 165.
Application Notes: Human rhinoviruses are known to cause a majority of upper respiratory tract infections, collectively known as common cold. This antibody binds a neutralizing epitope on the viral capsid protein VP2 of the human rhinovirus type 2. This antibody is capable of binding the denatured VP2 protein as well as recombinant VP2 expressed as a fusion protein in E. coli in Western blots (PMID: 2434607). This antibody is also capable of binding peptide 'VKAETRLNPDLQPTEC' in an indirect ELISA (PMID: 2822846, 1703215). This antibody can also immunoprecipitate HRV-2 virus (PMID: 1703215). This antibody was also used in the study of formation of distinct functional membranes after the human rhinovirus infection using immunohistochemistry (PMID: 17595532).
